Do calls made with the Manual Dial function count towards talk time and dial tics?

Not in the beta report_tool. However in the Tic Sheet in the dialer and in the Dialer Usage Report in the Reports tab the Manual Dials are reported.
However, if you want to be sure to track the talk time and the dial tics, you'll want to put the phone number in a record, save the record, and then dial it using the Call Now link.
